The first gun ever depicted on-screen in a videogame belongs to the 1975 Japanese arcade game, Western Gun. The game, developed by Taito, recreates a classic Wild West shoot-out, with two cowboys trying to kill each other before running out of bullets. It was released in the USA under the more self-explanatory title of Gun Fight, also in 1975, and was also the first arcade game to use a microprocessor to power its action.
